Eurofins MWG Operon offers standard genes which are available in any size, with
moderate GC content (40-65%), without extensive repeats (> 20 bp) or critical
hairpin structures and without extensive homopolymer stretches (> 20 bp). Our
proprietary software GENEius provides unique features for gene optimisation.
Processing Standard Genes
1. Sequence Optimization (optional)
By using our proprietary software, GENEius, the supplied amino acid or DNA sequences
will be optimized, using our proprietary 4D™ Optimization process. Codons are optimized
for the host species, repeats or hairpin structures are avoided, and G's and C's
are equally distributed. GENEius is also available as free online tool for our customers.
2. Experimental Design
Overlapping oligonucleotides used to create the gene are designed by GENEius.
3. Oligonucleotide Synthesis
All oligonucleotides are synthesized at our in-house DNA synthesis facility.
4. Gene Synthesis
Synthetic DNA is assembled using overlapping oligonucleotides, employing LCR (Ligase
Chain Reaction) and PCR technologies.
5. Subcloning
The gene can be cloned into a standard vector or on request into a vector of your
choice.
6. Quality Control
All genes undergo double-strand DNA sequencing at our in-house sequencing facility,
to ensure that each gene has the proper sequence.
Delivery and Documentation
The synthetic DNA will be delivered as plasmid DNA (mini scale preparation). Higher
scales are available on request. You receive a quality assurance report, including
detailed project specifications, complete plasmid description, restriction map and
DNA sequence information. The following special delivery options are available upon request: bacteria stabs, wet plasmid DNA, dry or wet linearized plasmid DNA, and PCR products.
